摘要:cationiclipid-coatedmagneticnanoparticles(mps)associatedwithtransferrinwereevaluatedasgenetransfervectorsinthepresenceofastaticmagneticfield.mpswerepreparedbychemicalprecipitationandweresurface-coatedwithcationiclipids,composedofddab/soypc(60:40mole/mole).thesecationicmpswerethencombinedwithpolyethylenimine(pei)condensedplasmiddna,followedbytransferrin.theresultingmagneticelectrostaticcomplexesretainedrelativelycompactparticlesizeandshowedcompletednacondensation.theirtransfectionactivityinthepresenceofastaticmagneticfieldwasevaluatedbyluciferaseandgreenfluorescentprotein(gfp)reportergenes.themagneticcomplexesexhibitedupto300-foldhighertransfectionactivitycomparedtocommonlyusedcationicliposomesorcationicpolymercomplexes,basedonluciferaseassay.theenhancementintransfectionactivitywasmaximizedwhenthecellswereexposedtothevectorsforarelativelyshortperiodoftime(15min),orweretreatedinmediacontaining10%serum.incorporationoftransferrinfurtherimprovedtransfectionefficiencyofthecationicmps.however,whencellswereincubatedfor4hinserum-freemedia,magneticandnon-magneticvectorsshowedsimilartransfectionefficiencies.inconclusion,transferrin-associatedcationicmpsareexcellentgenetransfervectorsthatcanmediateveryrapidandefficientgenetransferinvitrointhepresenceofamagneticfield.
