摘要:directgenetransferintotherespiratorysystemcouldbecarriedoutforeithertherapeuticorimmunizationpurposes.herewedemonstratethatcellsinthelungcantakeupandexpressplasmiddnaencodingaluciferasereportergenewhetheritisadministeredinnakedformorformulatedwithcationicliposomes.dependingonthelipidused,thetransfectionefficiencywithliposome-formulateddnamaybehigher,thesameas,orlessthanthatwithpureplasmiddna.tetramethyltetraalkylspermineanalogswithalkylgroupsof16or18carbonsanddmrie/cholesterolformulationsprovedparticularlyeffective.similarresultsforreportergeneexpressioninthelungwereobtainedwhetherthedna(nakedorlipidformulated)wasadministeredbyindirect,noninvasiveintranasaldelivery(inhaledorinstilled)orbyinvasive,directintratrachealdelivery(injectedorviaacannula).reportergeneexpressionpeaksaround4days,thenfallsoffdramaticallyby9days.thedose-responseislinear,atleastupto100μgplasmiddna,suggestingbettertransfectionefficienciesmightberealizediftherewasnotavolumelimitation.foragivendoseofdna,thebestresultsareobtainedwhenthednaismixedwiththeminimumamountoflipidthatcancomplexitcompley.theseresultsarediscussedinthecontextofdirectgenetransferforeithergenetherapyordeliveryofamucosaldnavaccine.
