hymag抗体 人乙肝表面抗原抗体（包被）
操作步骤
1．切片常规脱蜡至水。如需抗原修复,可在此步后进行
⒉ 缓冲液洗 3min/2 次。
⒊ 为了降低内源性过氧化物酶造成的非特异性背景染色,将切片放在 hydrogen peroxide block 中孵育 10-15 分钟。
⒋ 缓冲液洗 5min/2 次。
⒌ 滴加 ultra v block , 在室温下孵育 5 分钟以封闭非特异性的背景染色。
（注：孵育不要超过 10 分钟,否则会导致特异性染色降低。如果一抗的稀释液中含有 5 － 10% 正常羊血清,这一步可以省略。）
⒍ 缓冲液洗 5min/2 次。
⒎ 滴加一抗工作液 37 ℃孵育 1 － 2 小时。（具体孵育时间和温度由试验者最终决定）
⒏ 缓冲液洗 5min/2 次。
9 ． 滴加 primary antibody enhancer（增强子）,在室温下孵育 20 分钟。
10 ．缓冲液洗 5min/2 次。
11 ．滴加 hrp polymer（酶标二抗） ,在室温下孵育 30 分钟。
（注：hrp polymer 对光敏感,应避免不必要的光暴露并储存在不透明的小瓶中。）
12 ．缓冲液洗 5min/2 次。
13 ．向 1ml dab plus substrate （或 aec plus substrate) 中滴加 1-2 滴 dab plus chromogen （或 aec plus chromogen）,混匀后滴加到切片上,孵育 3 － 15 分钟。（具体时间由染色深浅决定。）
14 ．自来水充分冲洗,复染,脱水,透明,封片。
